u is a float in the range of 0 to 1. So multiplying by 65535 would produce a floating point value. I do not know if it is being rounded or truncated to an integer. But I suspect that the IM developers have done whatever is correct to make it properly an integer.

It requires you to have actually installed Ghostscript onto your system where ImageMagick can find it. Be sure there are no install errors for Ghostscript.

The constant Fred give, 65535, is correct for Q16 IM. For portability across any Q-number IM, use QuantumRange. Good point. Those reversed files probably have tiff:polarity=min-is-white rather than polarity=min-is-black

Use -define quantum:polarity=min-is-black or -define quantum:polarity=min-is-white to toggle the photometric interpretation for a bilevel image.

You probably should specify what version of ImageMagick and what version of Windows and also version of MINGW. Were there any error messages when installing MINGW or ImageMagick. The page id [0], should be fine. I do not see that as a problem. I showed above that by removing the colon and still having spaces and [0] at the end worked fine on my Mac (unix) system. You just have to enclose the whole filename and [0] in double quotes as I showed above.

On my Mac, if I try to put a colon in a filename, the Mac will object. It says "Try using a name with fewer characters, or with no punctuation marks"

This works fine for me on IM 6.9.9.35 Q16 Mac OSX Sierra using double quotes.

Is filename : test2.pdf the name of your file including the spaces and colon?

Some OS do not allow colons in filenames.

Try using double quotes.

I do not use CR2 images, but try telling the input to consider it truly as CR2 by prefacing with CR2:
